学習や記憶を強化する物質,P7C3が発見された。脳の海馬という部分は学習と記憶に重要な役割を果たすが,P7C3はマウスの海馬で新たに生まれたニューロンが死滅するのを防いだ。海馬でニューロン生成を制御するある遺伝子は,人間で欠損すると学習障害が生じ,実験動物で不活性化すれば海馬の形成に悪影響が生じる。この遺伝子を不活性化したマウスにP7C3を投与したところ,海馬の構造と機能が正常化することが分かった。また,海馬のニューロン生成が減少した高齢のラットにP7C3を投与すると,新たなニューロンの生成と保護が促進され,学習・記憶能力も増進したという。(吉田素子)


UT Southwestern Medical Center researchers(1) have found a compound(2) that preserves(3) newly created brain(4) cells(5) and boosts(6) learning(7) and memory in an animal study.

The study of this compound, which appears in the July 9 issue of Cell, springs from(8) a $2.5 million National Institutes of Health(9) Director’s Pioneer Award to Dr. Steven McKnight, chairman of biochemistry(10) at UT Southwestern and senior author(11) of the study.

Over a three-year period, the research team led by Dr. McKnight and Dr. Andrew Pieper, assistant professor(12) of psychiatry(13) and biochemistry at UT Southwestern, screened(14) 1,000 individual molecules(15) to see which ones might enhance(16) the production of neurons(17) in the adult mouse(18) hippocampus(19), a region of the brain critical to learning and memory. The scientists found that one of the compounds, called P7C3, achieved this by protecting newborn neurons from dying.

The researchers then administered(20) P7C3 to “knockout(21)” mice lacking a gene(22) that controls the generation of new neurons in the hippocampus. Humans who lack this gene have a variety of(23) learning disabilities(24), and the “knockout” rodents(25) show related abnormalities(26) as well as a poorly formed hippocampus. When the “knockout” mouse received P7C3, however, normal structure and function of the hippocampus were restored.

In elderly rats(27), which characteristically(28) show a decline in the birth and formation of hippocampal neurons, the researchers found that P7C3 increased both the birth and survival(29) of new neurons, and the memory and learning capability(30) of the aged rats.

“It’s been a wonderful experience,” Dr. Pieper said. “At first there was a lot of doubt, because we could have gone through(31) the whole screen and found nothing.”
